讲座(一):
题目:Design of an artificial membrane traffic system for molecular communication

Abstract: In biological systems, membrane trafficking plays important roles in inter- and intracellular transport of bioactive molecules and signals. The detailed mechanism of the membrane traffic process has been essentially clarified in the biochemical and biophysical viewpoint, however, artificial membrane trafficking has not been achieved up to the present time. Recently we developed an artificial membrane traffic system by employing lipid bilayer vesicles as a molecular sender, a molecular receiver, and molecular capsules. We observed budding and fission of the molecular capsules from a molecular sender triggered by a chemical signal. In the vesicular system equipped with the gemini peptide lipids as molecular switches, selective sending and receiving of the molecular capsules from a molecular sender to a molecular receiver were achieved by using a combination of photonic and ionic signals. In addition, fusion of the molecular capsules attached on a molecular receiver successfully proceeded by a molecular input signal. The present membrane trafficking would be utilized as a key process to construct a molecular communication system on artificial cell membranes.

题目:Metallosomes: metal-coated liposomes as a new class of artificial cell membranes

Abstract: Structural and functional hybridization of a biomolecule with another chemical component has been often adopted for design of intelligent bio-inspired materials. Along this line, we have recently developed the cerasome, a liposomal membrane having a ceramic surface, as a novel organic-inorganic hybrid material. Owing to its extremely high morphological stability as compared with the conventional liposomes, the cerasome performs as an excellent biomembrane model. In order to break fresh ground for the cerasomes as the bio-inspired hybrid materials, here we designed and prepared the metallosome, a metal-coated liposome, by employing electroless plating technique toward the cerasomes. The electroless plating effectively proceeded on the cerasome surface having metal coordination sites to afford the metal-coated liposomal membrane. Thickness of the metal layer was tuned in nanometer scale by changing the reaction conditions of electroless plating. In a similar manner, the magnetic cerasome was prepared as novel artificial cell membranes capable of manipulation by magnetic field.

题目:Cerasome as an infusible and cell-friendly gene carrier

Abstract: Since the discovery of lipofection, cationic lipids have been widely used as transfection agents in gene delivery. Recently, we developed an excellent transfection system using a cationic cerasome as a gene carrier. We found that the cerasome was infusible. The monomeric cerasome complex of plasmid DNA in a viral size indeed exhibited a remarkable transfection performance, such as high activity, minimized toxicity and serum-compatibility, toward uterine HeLa and hepatic HepG2 cells. This was in marked contrast to the non-silylated reference lipid, which forms fused, huge particles with significantly lower activity, by a factor of 102-103 and exhibited more pronounced toxicity. A couple of potential generalities of the present cerasome strategies with respect to nucleic acids to be delivered and cationic lipids as carriers are worth mentioning. The cerasome-plasmid complexation is strong and efficient, even at a stoichiometric lipid/nucleotide ratio. In this context, the cerasome could also be used as a size-regulated carrier for diverse types of functional nucleic acids, such as aptamers and siRNAs.
 
 
专家简介:
Jun-ichi Kikuchi教授是纳米仿生材料研究方面世界知名学者。1982 年日本九州大学工学院部获博士学位后,先后在日本九州大学任助教、副教授、教授。1998年起在日本奈良先端科学技术大学院大学物质创成科学研究科组建仿生材料研究室,任研究室负责人。从事领域有生物有机化学、超分子化学、物理有机化学及仿生化学等, 先后在《Nature Protocol》, J. Am. Chem. Soc.》, Chem Rev 》,《Chem Comm》,《J. Organic Chem》,Tetrahedron》等国际著名期刊发表SCI 论文180余篇,著书12 部。
